It's really hard to choose because Dean has so many golden lines throughout the show! If I had to choose a few, here would be some:

well dean says so many that i upendo so i cant choose a favourite but ill just put a few that i like from him. This is from season 4 episode yellow fever when they go to see this old guy (sam and dean hand old guy their IDs) Dean: 'Those are real..... obviously.' (Sam nudges dean to tell him to shut up)
And from sam probably Sam: 'Dude, i am not using this ID.' Dean: 'Why not?' Sam: 'Cos it says bikini inspector on it.'
Thats actually one of my favourite funny scenes from Supernatural when he is talking to the receptionist dean says she probably wont even ask to see sams id but she actually does and dean's fake laugh is hilarious and sams face so funny lol
